Description

Mono- and disaccharides can be determined using High Performance Anion Exchange Chromatography (HPAEC) with Pulsed Amperometric Detection (PAD).

This method was developed as an alternative to the enzymatic method (GS8-12) for the determination of mannitol as an indicator of dextran-formation in beet processing.

Other mono- and disaccharides such as glucose, fructose, sucrose or raffinose can be estimated from the same chromatograms.

Since amino acids interfere in the HPAEC analysis of sugars, in the case of samples containing noticeable amino acid contents, a treatment for the removal of amino acids should be used.

The method is applicable to beet brei extracts and beet juice samples.

The precision for the determination of Mannitol in Raw Juice was determined in a collaborative study in 2011:

For mannitol contents in raw juice in the range of 30 mg/L to 200 mg/L the mean repeatability limit (P = 95%) has been estimated to be 11% and reproducibility limit (P = 95%) has been estimated to be 26% of the determined mass concentration value.
